I'd think you a brave soul if you did. Sure you can boot it and run it, but
think about it. SpinRite assumes it has exclusive access to the drive, and
is modifying the drive at the lowest possible level, so if the host OS does
anything, that could make a mess. Steve Gibson says "holy smoke, please
don't do that", so I'll leave it to someone else to repair a drive under a
VM.

Spinrite 6 (w/ lowest-common-denominator drive-access params) ran without
complaints in VMware Player 2.0 build 45731. Don't have a sacrificial
drive, so I didn't test that. Do you know of a configuration where it
fails?
